β-Arbutin stands out with its chemical structure as a glucoside of hydroquinone, where a glucose molecule is linked to hydroquinone through a β-glycosidic bond. This structure makes it more stable than α-arbutin and safer than regular hydroquinone, which is known to have cytotoxic effects on skin cells.
β-Arbutin has a high purity of 99.5% and a specific rotation of –64° to –68°, which is characteristic of the true β-form structure. The product is highly stable, with moisture loss not exceeding 0.5%, and has a distinct melting point of 199–201°C, confirming its purity and quality. Importantly, it contains very low levels of free hydroquinone, not more than 10 ppm, ensuring safety for cosmetic use.

Chemical Structure: β-Arbutin has the molecular formula C₁₂H₁₆O₇ and a molecular weight of 272.25. It is a glucoside of hydroquinone, consisting of one glucose molecule linked to hydroquinone through a β-glycosidic bond at the C-1 position of the glucose. This β-form structure is more stable than the α-form and has a specific rotation of –64° to –68°, which is a characteristic parameter confirming the purity of β-Arbutin.

Mechanism of Action:
Tyrosinase Inhibition
β-Arbutin primarily works by inhibiting tyrosinase, the key enzyme involved in melanin production (melanogenesis). Tyrosinase converts L-tyrosine to L-DOPA and subsequently to dopaquinone, which are early steps in melanin synthesis. β-Arbutin binds to the enzyme’s active site, which contains copper ions (Cu²⁺), thereby reducing the enzyme’s activity and decreasing melanin formation.Competitive Inhibition
Because the hydroquinone portion of β-Arbutin structurally resembles L-tyrosine, the natural substrate of tyrosinase, β-Arbutin can compete with L-tyrosine for binding to the enzyme. Research Study
A study conducted by the Department of Dermatology, Kyoto University compared the efficacy and safety of β-Arbutin with other skin-brightening agents such as α-arbutin and hydroquinone. The results showed that β-Arbutin is significantly more stable than α-arbutin, particularly under conditions involving light and heat, and is considerably safer than hydroquinone, as it does not cause skin irritation or cytotoxicity.
In vitro tests demonstrated that β-Arbutin effectively inhibits tyrosinase, with a low IC₅₀ value, indicating strong inhibitory activity. Studies on melanocyte cells further confirmed that β-Arbutin can significantly reduce melanin production without adversely affecting cell growth. Additionally, the presence of a β-glycosidic bond in β-Arbutin allows for the slow and controlled release of hydroquinone, enhancing its skin-brightening efficacy while minimizing the risk of side effects.
